纤维素酶-微波联合提取蜈蚣草黄酮及其抗氧化研究 被引量:4

摘要 目的探索纤维素酶-微波联合提取蜈蚣草黄酮的最佳工艺条件及对其抗氧化性进行研究。方法在单因素的基础上进行正交实验,数据经SPSS软件分析。以黄酮提取物对超氧自由基的清除率作为指标分析其抗氧化性。结果纤维素酶-微波联合提取蜈蚣草黄酮的最佳工艺条件为:料液比1∶20,微波功率800 W,压强0.3 MPa条件下处理2 min后,再进行纤维素酶解,于65℃,pH6.5条件酶解4h(酶用量0.11%)。此条件下,总黄酮提取率达到14.44%。蜈蚣草黄酮提取液对超氧自由基有良好的清除率,其IC50为3.45 mg/ml。结论相比单一的微波法、纤维素酶解法,纤维素酶-微波联合提取蜈蚣草黄酮的最大提取率分别高出20%和10%,且其黄酮提取物有良好的抗氧化性,有很好的开发前景。 Objective To obtain the optimal extraction conditions of flavonoids from Pteris vittata using cellulase-microwave method.Methods Orthogonal experiments was used on the basis of the single factor experiments.The data was analyzed by SPSS.The scavenging rate against superoxide radical was used as an indicator to analyze the antioxidant activity of the flavonoids extract.Results The results showed that the optimum extraction conditions was as follows: material-liquid ratio 1∶20,microwave time 2 min,microwave power 800 w,microwave pressure 0.3 MPa,cellulose dosage 0.11%,cellulase hydrolysis temperature 65℃,pH6.5,hydrolysis time 4 h.Under these conditions,the extraction rate of flavonoids from Pteris vittata reached 14.44%.Flavonoids from Pteris vittata had a good scavenging rate against superoxide radical,and the IC50 was 3.45 mg/ml.Conclusion Compared to the single microwave method,the extraction rate increased by 20%,compared to the single cellulase hydrolysis method,the extraction rate increased by 10%,and the flavonoids extract from Pteris vittata has good antioxidant activity.
