摘要
目的:观察大麻素CB1、CB2受体在非酒精性脂肪性肝病(NAFLD)大鼠肝组织中的动态表达,探讨大麻素受体在大鼠NAFLD发病中的作用。方法:利用高脂饮食建立NAFLD大鼠模型,分别在第8、16、24周处死大鼠,HE染色观察肝脏脂肪变、炎症活动和纤维化程度,用免疫组化染色方法观察CB1、CB2受体在NAFLD不同阶段肝组织中的表达。结果:实验大鼠8周呈单纯性脂肪肝改变,16周肝组织弥漫性脂肪变,伴炎性细胞浸润,24周肝脏脂肪肝仍存在,但炎症程度有所减轻,出现程度不同的纤维化。正常大鼠肝脏CB1,CB2受体无明显表达,而在实验组大鼠中的表达均显著增加,CB1受体在第8、24周的表达强于CB2受体。结论:CB1、CB2受体在NAFLD大鼠肝组织中的表达明显增加,提示CB1、CB2受体在肝脏病变过程中发挥重要的作用。
Objective: To investigate the dynamic expressions of endocannabinoid receptors(CB1,CB2) in the livers of rats with non-alcoholic fatty liver disease(NAFLD) and explore the role of endocannabinoid receptors in the pathogenesis of NAFLD in rats.Methods: An experimental rat model of NAFLD was established by feeding rats with high fat diet.The rats were euthanized at the 8,16 and 24 weeks.Hepatic steatosis,inflammation activity and fibrosis degree were observed by routine HE staining.The expression of CB1 and CB2 in the livers were determined by immunohistochemistry at different NAFLD stages.Results: The experimental rats exhibited simple fatty 1iver at 8 weeks,diffuse hepatic steatosis with inflammatory cell infiltration at 16 weeks,and at 24 weeks hepatic steatosis was still present while the degree of inflammation reduced and with different grade of fibrosis.CB1 and CB2 were not expressed in the livers of normal rats,however,the expressions of CB1 and CB2 were significantly increased in the livers of experimental rats.The expression of CB1 was higher than that of CB2 at 8 and 24 weeks.Conclusion: The expression of CB1 and CB2 is significantly increased in the livers of rats with NAFLD,and may play an important role in the pathogenesis of NAFLD.
