摘要
目的探讨血清B型利钠肽(BNP)结合受体利钠肽受体A(NPR-A)和利钠肽受体C(NPR-C)与心力衰竭的关系。方法选择雄性Wistar大鼠78只,随机分为6组:正常4周组(N4组)、假手术4周组(F4组)、手术4周组(T4组)、正常12周组(N12组)、假手术12周组(F12组)、手术12周组(T12组),每组13只大鼠。采用腹主动脉缩窄法建立大鼠慢性心力衰竭模型。分别在第4、12周检测LVEF、左心室短轴缩短率(FS)、腹主动脉内径、心脏重量指数、病理切片HE染色观察心肌细胞形态学改变、RT-PCR检测左心室NPR-A、NPR-C mRNA的表达情况。结果与T12组比较,N4、F4、T4、N12、F12组大鼠LVFF、FS明显上升(P<0.05);与T4组比较,N4、F4、N12、F12组大鼠LVEF、FS明显上升(P<0.05),N4、F4、N12、F12、T12组大鼠NPR-A mRNA表达明显下调(P<0.05);与T4、T12组比较,N4、F4、N12、F12组大鼠NPR-C mRNA表达明显下调(P<0.05)。结论心脏NPR-AmRNA表达只在心力衰竭早期上调,而心脏NPR-C mRNA的表达在心力衰竭早期及晚期均上调。
Objective To study the relationship among BNP receptors natriuretic peptide receptor- A(NPR-A) ,natriuretie peptide receptor-C(NPR-C) and HF. Method Seventy-eight male healthy Wistar rats were randomized into 6 groups as follows:normal group aged 4 weeks(N4),sham-op- erated group aged 4 weeks(F4),aortic-banded group aged 4 weeks(T4),normal group aged 12 weeks(N12), sham-operated group aged 12 weeks (F12), aortic-banded group aged 12 weeks (T12). Chronic overload HF model was established by abdominal aortic constriction. Color Doppler ultrasonography was performed to measure the LVEF,fraction shortening and abdominal aorta inside diameter. The heart weight index was determined at the time of rat execution. Pathological examination of the left ventricle showed the morphological changes of myocardium cells. The expression of left ventricle cardiac NPR-A and NPR-C mRNA were determined by RT-PCR. Results Both EF and FS decreased dramatically in T4 group(P〈0.05), and decreased further more in T12 group than those of their corresponding N- and F-groups(P〈0.05). The heart weight index increased in T4 group(P〈0.05) ,and dramatically in T12 group(P〈0. 05). Myocardial cells arranged in perfect order, and no inflammatory cells infiltration existed in N4, N12,F4 and F12 groups. The myocardium fracture, fibronecrosis, inflammatory cells infiltration were shown in T4 group,and become more serious in T12 group than T4 group. Cardiac NPR-A increased in T4 group(P(0.05) ;while cardiac NPR-C mRNA increased both in T4 group and T12 group(P〈0.05). Conclusion The expression of cardiac NPR-A is upregulated only at early stage of HF,while cardiac NPR-C increases during the entire process of HF.
