摘要
目的探讨不同的低氧浓度对离体培养胎鼠脑皮质神经干细胞(NSCs)的增殖与凋亡的影响。方法体外不同低氧条件下(3%O2、5%O2、10%O2)培养胎鼠皮质来源的细胞,实验组分别分为低氧1、3、5 d组,常氧组为对照。免疫细胞化学染色鉴定NSCs,BrdU掺入法检测实验组NSCs的增殖;流式细胞仪检测实验组NSCs的凋亡。结果 3%O2低氧1d组NSCs凋亡率高于对照组,低氧5 d组NSCs的增殖率才明显升高;5%O2与10%O2组NSCs增殖率随着低氧天数的增加而逐渐升高,而细胞的凋亡率则随着低氧天数的增加而逐渐降低。结论低氧程度可影响NSCs的体外增殖与凋亡,且影响程度与低氧剂量和低氧时间有一定的量效关系。
Objective To observe the proliferation and apoptosis of cortical neural stem cells(NSCs) from embryonic mice under the different hypoxic concentrations in vitro.Methods NSCs from brain cortex of embryonic mice were cultured in different hypoxic concentrations(3% O2,5% O2 and 10% O2) in vitro,and all experimental groups were divided into hypoxia for 1,3 and 5 days group respectively,the normoxic group was as control.Immunocytochemical techniques were used to identify stem cells,Brdu incorporation was used to detect the proliferation of NSCs of experimental groups,flow cytometry was used to detect the apoptosis of NSCs of experimental groups.Results The apoptosis rate of 3% O2 hypoxia for 1d group was higher than that of control group,the cell proliferation rate of hypoxia for 5 d group had significantly increased;cell proliferation rate of group 5% O2 and group 10% O2 were increased with postponed time,but the apoptosis rate was gradually decreased with increased the number of days in hypoxia.Conclusion The degree of hypoxia can affect on the the proliferation and apoptosis of NSCs,and the effect had a certain dose-effect relationship on hypoxia dose and time.
