摘要 以苋菜种子为材料,进行苋菜离体繁殖研究.结果表明:苋菜种子经过75%酒精消毒1 min,再用升汞消毒8 min,在MS培养基上的萌发率可达87%以上;从无菌苗切取带2片子叶的下胚轴,在MS+3.0 mg.L-1 6-BA+0.2 mg.L-1 NAA培养基上诱导不定芽的效果最好;不定芽在MS+3.0 mg.L-1 6-BA+1.0 mg.L-1 IAA培养基上的增殖效果最好,增殖系数可达13.02;试管苗在1/2MS+0.2 mg.L-1 IBA培养基上的生根效果较好,生根率可达90.35%;在适宜的温度下,试管苗移栽到菜园土上的成活率可达92.5%以上. Seeds were used to study in vitro propagation of amaranth(Amaranth mangostanus L.).The results showed that the germination rate of seeds reached 87% or higher in the MS medium after they were disinfected with in 75% alcohol 1min and in 0.1% mercuric chloride 8 min.The best medium for adventitious bud induction of hypocotyl with two cotyledons was MS with addition of 3.0 mg·L-1 6-BA,0.2 mg·L-1 NAA,for propagation was MS with addition of 3.0 mg·L-1 6-BA,1.0 mg·L-1 IAA,by which the proliferation coefficient was up to 13.02,for rooting was 1/2MS with addition of 0.2 mg·L-1 IBA,by which the rooting rate was 90.35%.When the plantlets were transferred into the garden soil,the survival rate was up to 92.5% at fit temperature.
