摘要
目的:基于方证相应学说,运用蛋白质组学技术探寻肾阴虚证的标志蛋白,以期在蛋白质表达水平找到肾阴虚证的物质基础,进而阐述肾阴虚证的发生机理。方法:将昆明系雄性小鼠随机分为老年肾虚证模型组、左归饮组与青年正常对照组3组,对其肝、肾、睾丸组织蛋白质提取液及血清分别进行蛋白质双向电泳并对电泳图谱进行分析,找出左归饮调节纠正的蛋白质进行质谱分析鉴定和Wstern-blot验证。结果:找出左归饮调节纠正的蛋白质点24个,质谱鉴定出20个蛋白质。结论:找到并鉴定了一组肾阴虚证标志蛋白质,根据蛋白质的功能在蛋白质表达水平初步探讨了肾阴虚证的发生机理。
Objective:To find specific proteins of kidney-yin deficiency syndrome by using proteomic technology based upon the correspondence of prescription and syndrome,so as to explore the essence of kidney-yin deficiency syndrome on the expression level of protein.Meanwhile,the occurrence mechanism of kidney-yin deficiency symptoms is explained.Methods: Kunming male mice were randomly divided into three groups as follows: old kidney deficiency model group,Zuoguiyin group and youth normal group.The proteins of liver 、kidney、testis tissue and serum were separated and analysed by two-dimensional gel(2-DE) to find the proteins regulated by Zuoguiyin.The mass spectrograph was used to identify these proteins.Results: 24differentially expressed protein spots were detected,and 20 protein spots were identify.Conclusion: The project screens and identifies a group of specific proteins of kidney-yin deficiency symptoms.We preliminarily explore occurrence mechanism of kidney-yin deficiency symptoms on the protein expression level.
