视神经横断后生长抑制因子Nogo及其受体NgR mRNA表达的变化及意义 被引量:1

摘要 观察大鼠视神经横断后Nogo-A及其受体NgR mRNA表达的变化。将36只正常成年SD大鼠随机分为三组,视神经横断7d组、14d组和正常对照组,每组12只。分别提取视神经组织,使用实时荧光定量聚合酶链式反应法定量分析Nogo-A及NgR mRNA表达量的变化。实验结果显示,在大鼠的视神经组织中存在Nogo-A及其受体NgR基因的表达。与对照组相比,视神经横断7d后Nogo-A表达显著减少(P<0.05);视神经横断14d后,其表达较横断7d组显著增加(P<0.05),但仍明显低于对照组(P<0.05)。NgR在视神经横断7d后与对照组相比,表达显著减少(P<0.05),视神经横断14d后,其表达较横断7d组无显著差异(P>0.05)。视神经完全横断伤后Nogo-A mRNA表达先下降再升高,NgR mRNA表达下降并基本保持在同一水平。 To investigate the changes of Nogo-A and NgR mRNA expression in axotomic optic nerves of rat,36 adult normal SD rats were randomly divided into 3 groups: the normal control group,bilateral optic nerve transection treated 7 days later group and 14 days later group.There were 12 rats in each group.After tissue preparation,the mRNA levels were detected by real-time fluorescent quantitation polymerase chain reaction.The results showed that Nogo-A and NgR mRNA were expressed in the rat optic nerve.The level of the expression of Nogo-A mRNA in bilateral optic nerve transection treated 7 days later group was significantly lower than control group(P〈0.05);the level in 14 days later group was significantly higher than 7 days later group(P〈0.05),but still notably lower than control group(P〈0.05).The level of the expression of NgR mRNA in bilateral optic nerve transection treated 7 days was significantly lower later group than control group(P〈0.05),and there was not significant different between 14 days later group and 7 days later group(P〉0.05).After bilateral optic nerve transection treatment,the level of the expression of Nogo-A mRNA is decreased at first and then increased,and NgR mRNA expressed in a nearly constant lower level than normal.
