摘要
利用β-葡萄糖苷酶研究了黄芩苷酶解的影响因素及其动力学特征参数,以酶解率为指标考察了黄芩苷酶解的影响因素,并由反应速率计算了酶催化动力学参数rmax和Km值,实验得出较优的酶解条件为:黄芩苷∶酶液为0.1g∶20mL,pH 4.6,55℃搅拌1.5h,酶解率达70%左右,且Km为5.03g.L-1,rmax为0.216g.L-1.min-1.黄芩苷与β-葡萄糖苷酶亲和力强,酶解反应较易进行,在适宜条件下能得到很好的酶解效果.因此,进一步优化酶解条件有望获得更具实用性的酶解工艺.
In order to study factors and enzyme kinetic parameters of the baicalin′s enzymolysis by the β-glucosidase,to study the enzymolysis factors with the enzymolysis rate as an indicator and calculate the kinetic parameters rmax and Km by the enzymatic reaction rate.The better enzymolysis conditions are as follows: baicalin 0.1 g,enzyme solution 20 mL,stir the mixed solution for 1.5 h at the conditions of pH 4.6,55 ℃,the enzymolysis rate is up to 70%,Km is 5.03 g·L-1,rmax is 0.216 g·L-1·min-1,baicalin and β-glucosidase have a strong affinity.The enzymatic reaction is feasible and the effective under optimum conditions.Therefore,further optimization of the enzymolysis conditions is expected to be more pratical enzymatic process.
