墨兰根状茎增殖及生物反应器培养的可行性研究 被引量:4

摘要 [目的]探讨6-BA浓度对墨兰根状茎增殖的影响和反应器培养墨兰根状茎的可行性,为墨兰种苗生产中大量培养繁殖材料提供理论依据。[方法]以奇花墨兰组培根状茎为外植体,研究了6-BA浓度对其根状茎增殖的影响和根状茎生长动态,比较了在固体培养基和液态培养基振荡和生物反应器培养条件下根状茎增殖效果。[结果]当培养基中6-BA浓度为2.0 mg/L时,墨兰根状茎生长良好,增殖效果显著。对根状茎的增殖生长过程进行显微镜观察,结果发现接种后10 d起可见新的根状茎分化,接种后10~20 d根状茎数增加,接种后20~40 d根状茎数不再增加,但已分化的根状茎不断增大。比较不同培养方法对根状茎增殖生长的影响发现,在生物反应器中根状茎增殖效果显著好于固态培养基培养和液态培养基振荡培养。[结论]6-BA2.0 mg/L有利于墨兰根状茎增殖,利用生物反应器大量培养根状茎是墨兰种苗工业化生产中可利用的一种有效方法。 [Objective] The aim was to explore the effect of 6-BA concentrations on rhizome proliferation of Cymbidium sinense and feasibility of rhizome culture in bioreactor for providing mass propagules during industrial seedling production.[Method] The rhizomes of C.sinense cv.’Qihua’ in vitro were used as experimental material to investigate the effect of 6-BA concentrations on rhizome proliferation and compare different methods of culturing rhizome as well as solid culture,shake culture,and bioreactor culture.[Result] 2.0 mg/L 6-BA was benefit to rhizome proliferation and growth,the impact of proliferation was significant.New rhizomes formed from explants formed after 10 days of culture,and the formed rhizome numbers increased at 10-20 days;at 20-40 days,the new rhizomes did not differentiate but rhizome elongation was found.Rhizome proliferation and growth was promoted by bioreactor culture,20.6 of rhizomes per explant formed in bioreactor while 13.3 and 5.8 rhizomes per explants were harvested in shake and solid culture,respectively.[Conclusion] 2.0 mg/L 6-BA was favorable for rhizome proliferation in vitro and using bioreactor to mass produce rhizomes is an efficient way for industrial seedling production of C.sinense.
