摘要
交联酶聚集体(CLEAs)是一种新型的无载体固定化方法。本研究利用该法对来源于西梅籽的β-葡萄糖苷酶进行了固定化研究。优化了制备交联β-葡萄糖苷酶聚集体的条件,并探讨其在丙基β-D-葡萄糖苷合成中的应用。研究发现,最适的沉降剂及其比例、最适交联剂(戊二醛)浓度以及交联时间分别为正丙醇、2/1(V/V)、10 mmol/L和2 h。在最适条件下,正丙醇-CLEAs的活力回收达84%。该固定化酶催化丙基β-D-葡萄糖苷合成反应96 h后,收率为60%。
Cross-linked enzyme aggregates (CLEAs) technique is a novel cartier-free immobilization method. In the present work, fl-glucosidase from prune (Prunus domestica) seeds was immobilized by this method. The preparation conditions of CLEAs were optimized, and the synthesis of propyl β-D-glucopyranoside catalyzed by this immobilized enzyme was investigated. The optimum precipitant and its concentration, cross-linking agent (glutaraldehyde) concentration, cross-linking time were n-propanol, 2/1 (V/T), 10 mmol/L and 2 h, respectively. Under the optimum conditions, the activity recovery of n-propanol-CLEAs reached 84%. In the synthesis of propyl β-D-glucopyranoside catalyzed by the immobilized enzyme, the yield of the desired product was 60% after 96 h.
