摘要
目的:观察暖心胶囊对心衰大鼠心肌肌浆网Ca2+-ATP酶、血浆血管紧张素Ⅱ(AngⅡ)、醛固酮(ALD)以及血流动力学的影响。方法:将91只雄性SD大鼠随机分为正常对照组,阿霉素(ADR)心衰模型组,暖心胶囊低、中、高剂量组,卡托普利组以及心宝丸组,每组13只,腹腔注射ADR建立心衰大鼠模型,分别给予相应药物灌胃观察6周,行Ca2+-ATP酶、血浆AngⅡ、血浆ALD以及血流动力学检测。结果:暖心胶囊中、高剂量组可改善ADR心衰大鼠心脏的收缩和舒张功能,提高Ca2+-ATP酶活性,降低血浆AngⅡ、ALD含量(P<0.05,P<0.01)。结论:暖心胶囊可以对抗ADR心衰大鼠心功能的下降,其机制可能与提高Ca2+-ATP酶活性,下调血浆AngⅡ、ALD含量有关。
Objective: To study the effect of Nuanxin Capsule (NC) on sarcoplasmic reticulum Ca2+-ATPase, plasma angiotensin Ⅱ (Ang Ⅱ) and aldosterone (ALD), hemodynamic parameters in heart failure rats. Methods: SD rat models of heart failure rats were established by intraperitoneal injection of adriamycin(ARD). Rats were randomly divided into 7 groups: normal control group, ARD group, low-, moderate- and high-dose NC groups, captopril group and Xinbao Pills group, 13 rats in each group. After 6 weeks, hemodynamic parameters were detected, Ca2+-ATPase activity was measured, and the plasma concentrations of Ang II and ALD were determined. Results: The systolic and diastolic function in ADR-induced heart failure rats was improved, the activities of Ca2+-ATPase were increased, and the plasma concentrations of Ang II and ALD were reduced in moderate- and high-dose NC groups(P 〈 0.05 or P 〈 0.01). Conclusion: Nuanxin Capsule can counteract the impairment of heart function in ADR-induced heart failure rats, and its possible mechanism is related with the increase of activities of Ca2+-ATPase and the decrease of the plasma concentrations of Ang II and ALD.
