摘要
目的:研究牛膝多糖(ABPS)对人树突状细胞(DC)的功能的影响,为评价牛膝多糖对免疫系统功能的影响提供实验依据。方法:用浓度分别为0.1mmol/L、0.05mmol/L、0.025mmol/L的牛膝多糖处理成熟DC细胞72h,以CCK-8试剂检测各组DC的同种混合淋巴细胞反应(MLR)能力的差异;以ELISA双抗体夹心法检测各组DC分泌细胞因子IL-12p70、IL-17、TNF-α功能的差异。结果:与未处理组相比,各浓度牛膝处理组DC的同种MLR能力明显增强(p<0.05),尤其以0.1mmol/L处理组为甚。各浓度牛膝处理组DC的IL-12p70、IL-17、TNF-α的分泌能力有显著提高。结论:在本实验条件下,牛膝多糖对DC细胞因子的分泌及DC对同种T细胞的刺激能力具有一定的促进作用。
Objective:To study the effects induced by Achyranthes bidentata polysaccharides(APBS) on the functions of human dendritic cells(DC),which could offer the experimental evidence to estimate the influence on the functions of human immune system by APBS.Methods: Human DC were stimulated by APBS for 72hours,with the concentration for 0.1mmol/L,0.05mmol/L and 0.025 mmol/L,respectively.CCK-8 kit was adopted to examine the function of allo-mixed lymphocyte reaction(allo-MLR) of DC,and Enzyme Linked Immunosorbent Assay(ELISA) to identify the levels of IL-12p70,IL-17 and TNF-α secreted by DC.Results: Compared with the sham APBS stimulated group,after stimulated by APBS,the ability of DC allo-MLR in each APBS treatment group was increased obviously(p0.05),especially the group with the concentration of APBS for 0.1mmol/L.However,the secreted levels of IL-12p70,IL-17 and TNF-α of DC in each APBS treatment was increased obviously(p0.05).Conclusion:Our study showed that APBS stimulation could somewhat up-regulate the secreted levels of IL-12p70,IL-17 and TNF-α and stimulation ability to allo-T cells of human DC.
