摘要
目的探讨多发性硬化(MS)患者血清瘦素(leptin)水平的变化与外周血单个核细胞(PBMCs)分泌细胞因子水平的关系及其临床意义。方法采用放射免疫法检测25例活动期MS患者、22例缓解期MS患者和30例健康对照者的血清leptin水平,同时测定其PBMCs在外源性leptin干预下培养上清液IL-4、INF-γ水平。结果活动期MS患者血清lep—tin水平、PBMCs在leptin干预下培养上清液INF-γ、IL-4水平均高于缓解期MS患者和对照组(P〈0.01);经过糖皮质激素治疗后活动期MS患者血清leptin水平、PBMCs在leptin干预下培养上清液INF-γ水平明显降低(P〈0.01),而IL-4水平显著升高(P〈0.01)。结论活动期MS患者血清leptin水平升高,leptin能够影响MS患者PBMCs炎性细胞因子分泌,这可能与瘦素增加MS的易感性相关。
Objective To explore the change of serum leptin levels in patients with multiple sclerosis (MS) and evaluate the effect of recombinant human leptin (rhleptin) on eytokine production by peripheral blood mononuelear cells (PBMCs) from MS patients. Methods A quantitative radioimmunity methods according to the manufaetur, s protocol was adopted to detect serum leptin levels in 25 active MS patients ,22 remission MS patients and 30 normal, at the same time we incubated PBMCs from all samples with rhleptin, the superuate IL - 4 production was measured by radioimmunity methods, IFN - γ levels were determined using an enzyme -linkod immunosorbent assay(ELISA). Results The serum leptin level in active M.S patients increased (P 〈 0. 01 ). Significant decreased serum leptin level was observed after methylprednisolone treatment in active MS patients (P 〈 0. 01 ). The IFN -γ, IL -4 secretion in supernate of PBMCs cultured with rhleptin increased(P 〈0. 01 ) in active MS group. The IFN -γ dccreased, but the IL -4 increased after methylprednisolone treatment. Conclusions The serum leptin level is high in active MS patients and it is related to therapeutic effect. Leptin can affect on pro - and anti - inflammatory eytokine production by PBMCs collected from MS patients, may he this connected with leptin increase the susceptiveness of MS.
