摘要
目的探讨睡眠剥夺(SD)对老龄大鼠学习记忆的影响及可能机制。方法 90只雄性SD老龄大鼠随机分成对照组、模型组、SB203580干预组。改良多平台SD法建立SD模型。SD1 d、3 d、5 d光镜观察海马区神经细胞形态变化,免疫组化法和Western印迹法检测海马磷酸化P38MAPK蛋白和β淀粉样肽(Aβ)表达,水迷宫法测试动物学习记忆功能。结果与对照组比较,模型组中,随睡眠剥夺时间延长,SD老龄大鼠海马神经元结构损伤明显,磷酸化P38MAPK和Aβ阳性细胞数及表达增多,动物学习记忆能力减退;与模型组比较,SB203580组中脑组织形态损伤程度减轻、磷酸化P38MAPK和Aβ阳性细胞数及表达,动物学习记忆功能得到改善。结论睡眠剥夺可加重老龄大鼠学习记忆功能障碍,与P38MAPK信号过度活化、Aβ沉积有关。
Objective To investigate the effect of sleep deprivation(SD) on learning-memory in elderly rats.Methods Elderly male SD rats were divided into 3 groups: control,model,SB203580 treatment groups.SD models were established by the modified multiple platform methods.At 1 d,3 d,5 d after SD,morphological changes were observed with light microscopy.The expression p38MAPK phosphorylation and Aβ were detected by immunohistochemistry and Western-Blot.Learning-memory function was performed with Morris water maze.Results Compared with control group,neuronal morphous damaged obviously,the expression of p38MAPK phosphorylation and Aβ were increased,learning and memory function significantly were decreased with time of SD in model group(P 0.05).Compared with model group,the morphous injury,the p38MAPK phosphorylation and Aβ expressions were decreased obviously in SB203580 treatment group(P 0.05).Meanwhile,the ability of learning and memory function enhanced(P 0.05).Conclusions SD can aggravate learning-memory impairment in elderly rats,which is related to P38MAPK activation and Aβ expression after SD.
