二甲双胍对血管内皮细胞磷酸化腺苷酸活化蛋白激酶和一氧化氮含量的影响 被引量:1

摘要 目的观察二甲双胍(降血糖药)对人脐静脉内皮细胞(HUVEC)的磷酸化腺苷酸活化蛋白激酶(P-AMPK)表达和一氧化氮含量的影响。方法将体外培养的人脐静脉内皮细胞分为6组:①空白对照组(常规培养);②二甲双胍(Met)2 mmol·L-1组;③软脂酸(PA)300μmol·L-1组;④软脂酸300μmol·L-1+二甲双胍2 mmol·L-1组;⑤5-氨基咪唑-4-甲酰胺核糖核苷酸(AIC-AR)1 mmol·L-1组及⑥软脂酸300μmol·L-1+AICAR 1 mmol·L-1组,经孵育24 h后,收集细胞和培养上清液,用Western blot方法,测定各组的AMPK的表达;用硝酸盐还原法,观察各组培养上清液中的一氧化氮的含量。结果与空白对照组比较,Met组AMPK的磷酸化表达水平升高,培养上清液中NO生成增加。(Met+PA)组AMPK的磷酸化表达水平升高,NO生成增加。结论二甲双胍通过激活AMPK使NO生成增加,改善HUVECs的内皮功能,从而起到抗动脉粥样硬化的作用。 Objective To investigate whether metformin can improve the endothelial function through activating AMP-activated protein kinase(AMPK) in human umbilical vein endothelial cells(HUVEC).Methods HUVEC were cultured in vitro.The study was designated to 6 groups: ①control group(conventional culture);② metformin group(2 mmol·L-1);③ palmatic acid group(300 μmol·L-1);④metformin+palmatic acid group(2 mmol·L-1+300 μmol·L-1);⑤ 5-aminoimidazole-4-carboxamide-ribo-nucle-oside(AICAR) group(1 mmol·L-1);⑥ palmatic acid+AICAR group(300 μmol·L-1+1 mmol·L-1).After HUVEC were incubated for 24 hours,the expression of phosph AMP-activated protein kinase of human umbilical vein endothelial cells were evaluated with Western blot.The content of nitric oxide(NO) were measured with nitrate reductase.Results Compared with the control group,metformin could increase the expression of phospha AMPK,and increase the content of NO.Conclusion The endothelial function was improved by through activating the AMPK.
