摘要
利用18对微卫星引物测定云南省、西藏自治区4个地方特色鸡种(‘藏鸡’、‘勐腊茶花鸡’、‘贡山土鸡’、‘武定鸡’)的遗传多样性,对等位基因频率、等位基因数、有效等位基因数、群体杂合度和多态信息含量等遗传多样性参数进行了统计分析,并依据Nei氏DA遗传距离,采用NJ法和UPGMA法构建了系统聚类树.结果表明:18个微卫星座位均表现高度多态,4个鸡群体138个个体中共检测到244个等位基因,平均期望杂合度为0.727,平均多态信息含量为0.669,遗传距离最远的是‘藏鸡’与‘勐腊茶花鸡’(DA=0.400 3,Ds=0.567 5),最近的是‘贡山土鸡’与‘武定鸡’(DA=0.117 8,Ds=0.089 5).
The genetic diversity of 4 indigenous chicken breeds in Yunnan Province and Tibet was analyzed by 18 microsatellite markers.Allele frequency,number of observed alleles(Na),number of effective alleles(NEA),heterozygosity(H),and polymorphic information content(PIC) were calculated among other genetic diversity parameters.NJ and UPGMA phylogenetic trees were reconstructed using Nei's DA genetic distances.The results indicated that a total of 244 alleles were detected in 138 samples of the 4 populations,the estimates of average expected heterozygosity(He) and PIC per locus were 0.727 and 0.669.'Tibetan Chicken' and 'Menglachahua Chicken' had the farthest genetic distance(DA=0.400 3,Ds=0.567 5),'Gongshan Chicken' and 'Wuding Chicken' had the nearest genetic distances(DA =0.117 8,Ds=0.089 5).
