摘要
目的建立分析鼠疫耶尔森菌外膜蛋白抗原性的简便方法。方法采用DNAstarProtean和EMBOSS网络分析平台综合预测鼠疫耶尔森菌外膜蛋白YopE、YopH、YopT、YopJ、YopM的免疫原性,并通过蛋白质芯片技术对上述外膜蛋白的免疫原性进行验证。结果计算机综合预测分析显示外膜蛋白YopE抗原性最好,蛋白质芯片技术进一步证实YopE具有良好的免疫原性。结论软件预测结合蛋白质芯片检测的方法可用于筛选病原微生物抗原性蛋白。
Objective To establish a feasible method for studying antigenicity of the outer membrane proteins of Yersinia pestis(Yops).Methods Secondary structure,flexibility,hydrophilicity and antigenicity index of Yops were predicted by DNAStar Protean softeware and EMBOSS net platform.And immunogenicity of Yops was detected by protein chip.Results The prediction by computer was consistent with results from protein chip,proving YopE had the best immunogenicity.ConcIusion Precise computer prediction combined with protein chip screening is an effective research method for screening antigenic protein in pathogenic microorganism.
