摘要
【目的】观察新型透皮吸收中药贴剂穴位敷贴对哮喘小鼠相关转录因子的调控作用。【方法】采用随机对照分组法将60只BALB/c小鼠随机分为正常组、模型组和贴药组,除正常组外,其他两组均采用卵蛋白致敏与激发法造模,其中贴药组以新型透皮吸收中药贴剂敷贴小鼠肺俞、大椎、风门穴,疗程结束后分别应用双抗体夹心酶联免疫吸附(ELISA)法检测各组免疫球蛋白E(IgE)、白细胞介素4(IL-4)的含量;采用逆转录—聚合酶链反应(RT-PCR)法检测T-盒21基因(T-betmRNA)、GATA结合蛋白3基因(GATA-3 mRNA)表达。【结果】新型透皮吸收中药贴剂穴位敷贴可显著降低哮喘小鼠IgE、IL-4水平,抑制转录因子GATA-3的基因表达,同时增强转录因子T-bet的基因表达。【结论】穴位敷贴在体内可能主要通过GATA-3、T-bet等转录因子调节Th2细胞反应,从而达到治疗哮喘的目的。
Objective To observe the regulation of transcription-associated factors in asthmatic mice by neotype Chinese medicinal transdermal patch for point application.Methods Sixty mice were randomly divided into normal group,model group,and point application group.Except for the normal group,the mice in other two groups were sensitized and challenged with ovalbumin.Point application group was given application of neotype Chinese medicinal transdermal patch on points Feishu,Dazhui and Fengmen.After treatment,enzyme-linked immunosorbent assay(ELISA)was used to detect the levels of immunoglobulin E(IgE) and interleukin 4(IL-4),and reverse transcription polymerase chain reaction(RT-PCR) assay was used for the detection of mRNA expression of T-box expressed in T cells gene 21(T-bet) and GATA binding protein 3 gene(GATA-3).Results Compared with the model group,IgE and IL-4 levels were reduced,GATA-3 gene mRNA expression was inhibited,and T-bet gene mRNA expression was increased in point application group.Conclusion The therapeutic mechanism of point application for asthma is probably related with the regulation of transcription-associated factors of GATA-3 and T-bet in Th2 response.
