摘要
应用分子生态学方法,从大庆油田萨南开发区高台子油层样品(包括1个注入水样和8口油井产出液样组成的混合样)中提取总DNA,扩增其16S rDNA片段,克隆到大肠杆菌,建立16S rDNA克隆文库,研究细菌群落结构.结果表明:从注入水样的微生物得出22个分类操作单元(OUT),从油井产出液样的微生物得出9个分类操作单元(OUT),注入水样的微生物多样性明显高于油井产出液样的.对克隆测序的16S rDNA序列进行GenBank数据库比对和系统发育分析,注入水样的细菌属于γ变形菌纲(79.0%)、β变形菌纲(3.0%)和未培养微生物(18.0%);油井产出液样的细菌属于γ变形菌纲(87.7%)、δ变形菌纲(8.5%)和未培养微生物(15.1%).与石油烃降解和产生物表面活性剂相关的假单胞菌(Pseudomonas)在注入水文库和油井产出液文库中的比例分别为51.0%和87.7%.油藏微生物群落结构的研究可以为微生物采油技术提供基础资料.
To develop microbial enhanced oil recovery(MEOR) technology,bacterial diversity of Gaotaizi Reservoir in Sanan development area in Daqing Oilfield was investigated by molecular biology methods.One sample of injected water and one mixed sample including eight samples of produced water were analyzed.The genomic DNA was extracted,16S rDNA clone library was constructed by PCR amplification with universal primers,cloned in Escherichia coli and bacterial community structure was studied by the analysis of 16S rDNA clone library.Bacterial clones of injected water were classified into 22 different operational taxonomic units(OUTs),bacterial clones of produced water were only classified into 9 different OUTs.The result showed that the diversity of microbial communities in the injected water was more complicated than that in produced water.16S rDNA of each clone was sequenced and phylogenetic analysis revealed that the dominant microbes of the injected water belonged to Gammaproteobacteria(79.0%),Betaproteobacteria(3.0%),and the uncultured bacteria(18.0%);the dominant microbes of the produced water were composed of Gammaproteobacteria(87.7%),detaproteobacteria(8.5%) and the uncultured bacteria(15.1%).Pseudomonas related with hydrocarbon biodegradation and surfactant production accounted for 17.79 % and 6.02 % in the clone library of injected water and produced water,respectively.These results provide the basis information on microbial ecology in specific reservoirs for MEOR technology.
