摘要
目的:研究不同浓度Borrelidin对口腔颌面部静脉畸形模型小鼠脾脏及血液中VEGF的作用。方法:随机将45只8周龄SPF雄鼠,质量25~30g,作为NC组;慢性束缚应激模型制备成功的225只SPF受孕雌鼠,质量25~30g,随机分为5组:EX组、BO0.8组、BO1.8组、BO2.8组、BO3.8组,每组45只。每组又随机分3d、7d、14d组,每组15只。NC组正常饲养不做处理;EX组开腹后只做注射动作,BO各浓度组小鼠脾脏直接注射相应浓度的Borrelidin注射液。各组分别在3、7、14d3个时间点手术实施主动脉穿刺采血,ELISA法检测各组血样中VEGF的浓度。采血后用免疫组化(SABC)法检测小鼠脾脏组织VEGF的表达并行病理图像定量分析和统计学分析。结果:血液VEGF浓度:NC组仅有微量表达。EX组术后浓度呈持续高表达,与NC组有显著统计学差异。BO0.8、1.8、2.8、3.8组均可降低血液中VEGF的浓度。SABC图像定量分析结果显示VEGF表达变化的趋势同ELISA检测。结论:Borrelidin可有效降低静脉畸形模型小鼠脾脏VEGF的表达和血液VEGF的浓度。
Objective:To study effects of different concentrations of Borrelidin on VEGF in the blood and spleen of mice as venous malformation model of oromaxillo-facial region.Methods:45 SPF male mice,aged 8weeks and weighing 25-30g,are randomly chosen as NC group.225 SPF conceived female mice were prepared as chronic bondage stress model,which weighted 25-30g.They were randomly divided into 5 groups:EX group,BO0.8 group,BO1.8 group,BO2.8 group,BO3.8 group,45/ per group.Each subgroup was randomly divided into 3 days,7 days,14 days group,15/ per group.NC group was normally raised;EX group got nothing after laparotomy.Mice spleen of different concentrations of BO groups injected with corresponding concentrations of Borrelidin.Each group was taken blood by aortic piercing operation at 3 days,7days and 14 day respectively,and then the concentrations of VEGF in different blood samples were tested using ELISA.After taking blood,expressions of VEGF in splenic tissue were tested using immunohistochemical method(SABC) and processed with pathological image quantitative analysis.Results:Concentration of VEGF in blood:trace expression in NC group;constant high expression after operation in EX group and significant statistic difference with NC group;the concentrations of VEGF in blood decreased in BO0.8,1.8,2.8,3.8 groups.The changing trend of the expression of VEGF by immunohistochemical image quantitative analysis was same as that by ELISA test.Conclusion:Borrelidin can reduce effectively the expression of VEGF in mice spleen and concentrations of VEGF in blood of venous malformation model.
