摘要 目的:探讨GLUT4及瘦素在大鼠非酒精性脂肪性肝病(NAFLD)中的作用。方法:64只SD大鼠随机分成正常对照组(32只)和高脂饮食组(32只),每组又分为2周、4周、8周、12周四组,按时间点处死各组大鼠,测定大鼠肝组织中GLUT4的含量、血清瘦素含量、空腹血糖(FBG)、空腹血清胰岛素水平(FIns),观察肝组织病理改变。结果:高脂组大鼠4周时出现非酒精性脂肪性肝病(NAFLD),12周时呈现典型的脂肪性肝炎(NASH)表现,并出现肝纤维化。高脂组大鼠的GLUT4、血清瘦素、FBG、FIns、肝细胞脂肪变程度与对照组相比均有差异。血清瘦素、FBG、FIns与肝脂肪变程度呈正相关,GLUT4、与肝脂肪变程度呈负相关。结论:高脂组大鼠肝组织中的GLUT4与对照组大鼠相比明显减少,血清瘦素含量与对照组大鼠相比则明显增加;GLUT4及瘦素在NAFLD的形成过程中起重要作用。 Objective :To investigate the change and significance of GLUT4 and Leptin in liver homogenate and blood with nonalcoholic fatty liver disease in rats. Methods:Sixty four rats were randomly assigned to con- trol group and high fat diet group with 32 in each. Each group was divided into four subgroups: 2-- week, 4- week, 8-- week and 12-- week groups. The rats of all groups were killed at the right time. The liver GLUT4 level, blood Leptin level, FGP, FINS and IRI were detected, and then the histological changes were observed under light microscope. Results :The liver of rats in high fat diet group showed the changes of NAFLD at Week 4, and showed the NASH and liver fibrosis changes at week 12. Compared with the control group, the GLUT4, Leptin, FBG, FINS and the level of steatosis in high fat diet group showed significant changes. Lept- in, FGB and FINS had a positive correlation with the steatosis level but GLUT4 had a negative correlation with the level. Conclusion : GLUT4 in the rats of high fat diet group were reduced significantly compared with that of the control group. GLUT4 and leptin play an important role in the pathogenesis of NAFLD.
