摘要
目的建立一种简便、快速的方法测定人血浆中华法林的含量。方法采用ZORBAXEclipseXDB-C8色谱柱(150mm×4.6mm,5μm);流动相为0.01mol·L^-1磷酸二氢钾溶液-乙腈(55:45);流速为1.0mL·min^-1;检测波长为308nm。结果华法林保留时间约为6.2min,线性范围为248-3720μg·L^-1,r=0.9993,华法林的高、中、低质量浓度日间、日内RSD均在10%以下,回收率大于90%。结论该法简便、快速,结果准确,可用于临床上患者血浆中华法林药物浓度的监测。
Objective To establish a simple and fast method for determination of warfarin in human plasma. Methods RP-HPLC method was used with C8 column (150 mm×4.6 mm,5 μm) ,and 0.01 mol · L^-1KH2PO4-CH3CN solution (55 : 45) was used as mobile phase. The flow rate was 1.0 mL× min^-1. DAD was utilized at 308 nm. Results The retention time of warfarin was 6.2 min. The linear range of the standard curve was from 248 to 3 720 μg · L^-1 with a correlation coefficient of 0. 999 3. The average recovery was over 90 %. RSD of the high, middle and low concentrations were less than 10%. Conclusion The method is simple, fast, accurate and can be used to monitor warfarin plasma concentrations in clinical practice.
