移植后的Wistar大鼠骨髓间充质干细胞向神经元分化的实验研究 被引量:2

摘要 目的观察Wistar大鼠骨髓间充质干细胞(BMSCs)移植后在脊髓损伤区是否向神经元分化。方法首先用BrdU在体外标记BMSCs,将标记后的BMSCs分别由大鼠尾静脉及脊髓损伤局部移植到大鼠体内,于移植后10天、20天及30天灌杀大鼠,制作脊髓的冰冻切片并行免疫组织化学染色,在激光共聚焦显微镜下观察植入的BMSCs表达神经元特异性蛋白NSE的情况。结果移植术后10天2、0天及30天,损伤脊髓内可见BrdU/NSE双标记的细胞。结论 BMSCs移植后在脊髓损伤区内可见神经元特异性蛋白NSE,移植的BMSCs可能分化为神经元。 Objective To transplant Wistar rats' bone marrow mesenchymal stem cells(BMSCs) into the rats' spinal cord injury zone.observe the BMSCs Whether or not to differentiate into neurons.Methods First of all,marker BMSCs with BrdU in vitro,the BMSCs will be transplanted respectively by the rat tail vein and partial spinal cord injury in rats.By the 5th days,10th days,15th days after the transplantation,these rats are perfused to kill for the spinal cords.The spinal cords are maked into frozen section,which are examined under the laser scanning confocal microscope for the condition that the transplanted BMSCs express the neuron-specific protein-NSE.Results 10,20,30 days after transplantation,under the laser scanning confocal microscope,were observed BrdU / NSE double labeling cells.Conclusion The transplanted BMSCs can express the neuron-specific protein-NSE,which May differentiate into neurons.
