摘要
目的观察"芩部丹"中3种单体对巨噬细胞TLR2的调节作用,以探寻其抗结核的可能作用机制。方法取黄芩苷、对叶百部碱、丹参多酚酸盐单体作用于感染结核杆菌的U937单核巨噬细胞,分别采用RT-QPCR、FACS方法检测用药前后巨噬细胞表面TLR2的表达差异。结果与模型组相比,黄芩苷、对叶百部碱、丹参多酚酸盐均具有上调TLR2表达的作用。结论 "芩部丹"中3种单体抗结核作用可能通过纠正TLR2的表达而发挥作用。
Aim To observe the possible molecular mechanisms of Qibudan against tuberculosis infection. Methods U937 cells infected with H37Ra strain of Mycobacterium were treated with baicalin, tuberoste- monine and salvianolate. Then the expression of TLR2 was analysed by Realtime PCR and FACS. Results After infected with H37Ra, the expression of TLR2 in U937 cells was reduced significantly in mRNA level. Similar result was obtained protein expressionTLR2. When added baicalin, tuberostemonine and salvianolate, the mRNA was found and protein expres- sions of TLR2 were all increased to normal. Conclusion Qibudan can TLR2, which suggests one of the mechanisms infection. up- that regulate the expression of regulation of TLR2 can be of Qibudan against tuberculosis
