橙皮苷对酒精性脂肪肝大鼠脂质过氧化及TNF-α生成的影响 被引量:7

摘要 目的研究橙皮苷(HDN)对大鼠酒精性脂肪肝(AFLD)的防治作用并探讨其对脂质过氧化及肿瘤坏死因子α(TNF-α)生成的影响。方法以酒精联合脂肪乳剂灌胃,每天2次,连续6周,制备大鼠AFLD模型。于第6周末采血,检测血清丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)、总胆固醇(TC)、甘油三酯(TG)、TNF-α水平及肝脏超氧化物歧化酶(SOD)、丙二醛(MDA)含量,并行肝脏病理学检查。结果 HDN能明显减轻AFLD大鼠的肝脏脂肪变和炎症程度,明显降低AFLD大鼠肝脏指数和血清中ALT、AST、TC、TG、TNF-α水平,降低肝匀浆中MDA的含量,提高SOD的活力。结论 HDN对大鼠AFLD有一定的防治作用,其机制可能与抗脂质过氧化及降低血清TNF-α水平有关。 Objective To investigate the therapeutic effects of hesperidin(HDN) on alcoholic fatty liver disease(AFLD) in rats and to explore the role of HDN on lipid peroxidation and tumor necrosis factor-alpha(TNF-α).Methods In this study,the rat model of AFLD could be developed by intragastric alcohol and fat emulsion twice a day for six weeks.Then the levels of MDA and SOD in liver tissue,and ALT,AST,TC,TG,TNF-α levels in serum were measured.HE staining was used to evaluate the degree of AFLD.Results Liver histopathological examination showed that HDN could ameliorate steatosis and inflammation of hepatocytes.At the same time,HDN could not only reduce the liver index and ALT,AST,TC,TG,TNF-α in serum significantly,but also enhance SOD activity and reduce MDA contents in liver tissue obviously.Conclusion HDN has positively therapeutic effects on AFLD,which may be associated with its effects of anti-lipid peroxidation and reducing TNF-α in serum.
