摘要
目的:研究6,8-二-三氟甲基-7-乙酰氧基白杨素(dFMAChR)抑制荷人宫颈癌裸鼠移植瘤生长的可能机制。方法:建立人宫颈癌裸鼠移植瘤模型,通过免疫组织化学SP染色法观察dFMAChR对人宫颈癌移植瘤组织血管内皮生长因子(VEGF)、细胞增殖核抗原(PCNA)表达的影响。结果:免疫组织化学检测发现dFMAChR抑制人宫颈癌移植瘤组织VEGF和PCNA的表达,且呈剂量依赖性。结论:dFAMChR能有效抑制人宫颈癌裸鼠移植瘤细胞PCNA和VEGF蛋白表达,这可能是dFMAChR抑制人宫颈癌裸鼠移植瘤生长的分子机制之一。
Objective: To investigate the possible mechanism of the inhibition effect of 6,8-ditrifluoromethyl-7- acetoxychrysin (dFMAChR), a novel chrysin derivative, on growth of xenograft of human cervical cancer. Methotis: Human cervical carcinoma xenograft in nude mouse was established and the anti-cervical cancer effect of dFMAChR in vivo was evaluated. Expression of proliferating cell nuclear antigen (PCNA) and vascular endothelial growth factor (VEGF) in human cervical cancer xenografts were detected by SP immunohistochemistry. Results: Therapeutic trial of human cervical carcinoma xenograft in nude mouse indicated that dFMAChR significantly inhibited the growth of human cervical carcinoma xenografts in Balb/c-nu mice in a dose- and time-dependent manner. Analysis of the expression of VEGF and PCNA protein by SP immunohistochemistry showed that they can be downregulated by dFMAChR at higher concentration. Conclusions: The present study suggests that dFMAChR has a negative effect on the angiogenesis in tumor xenograft of human cervical cancer by down-regulating the expression of PCNA and VEGF.
