摘要
目的:观察机械牵张应变作用下,人牙周膜细胞(HPDLCs)内磷脂酶C-γ1(PLC-γ1)水平的变化。方法:通过原代和传代培养,获得性状稳定的人牙周膜细胞,使用动态机械应变细胞加载仪对细胞进行1%、10%和20%动态牵张应变加载。加载不同时间,通过流式细胞仪对细胞进行PLC-γ1水平检测。采用SPSS13.0软件包对数据进行方差分析。结果:在加载牵张应变初期,细胞内的PLC-γ1水平维持较低水平;随着牵张应变加载时间的延长,牙周膜细胞内的PLC-γ1水平逐渐升高。50min时,PLC-γ1水平达到各应变组的最高值(P<0.01)。随着加载时间的继续延长,60min时,各应变组的PLC-γ1水平都显著下降(P<0.01)。结论:牵张应变可引起细胞内PLC-γ1水平的变化。
PURPOSE: To investigate the effects of mechanical stretching on concentration of phospholipase C-γ1(PLC-γ1) in the human periodontal ligament cells(HPDLCs).METHODS: Primary human periodontal ligament cells were obtained and passaged.Then the cells were stretched by dynamic mechanical strain unit with 1%,10% and 20% strain at different time.PLC-γ1 was examined through flow cytometry.The data were analyzed by one-way ANOVA using SPSS13.0 software package.RESULTS: In early period of stretching,the expression of PLC-γ1 remained at a lower level.With the stretching time prolonging,the expression of PLC-γ1 increased gradually.The level of PLC-γ1 of each stretching group reached peak concentration at 50-minute(P〈0.01),then declined at 60-minute(P〈0.01).CONCLUSION: Stretching force induces the change of concentration of the PLC-γ1 of HPDLCs.Supported by National Natural Science Foundation of China(30900282),Shanghai Leading Academic Discipline Project(S30206,T0202),Research Fund of Science and Technology Commission of Shanghai Municipality(08411961500,10QA1404200),Natural Science Foundation of Shanghai Municipality(07ZR14070) and Research Fund of Shanghai Research Institute of Stomatology(200805).
