摘要
目的探讨Bmi-1 mRNA的表达在肝癌发病过程的作用。方法 Trizol方法常规提取液氮保存的肝癌组织、肝硬化组织及正常肝组织总RNA。将这些总RNA逆转录成cDNA后,利用实时荧光定量PCR的方法检测Bmi-1基因在这几种组织中的表达情况,分析其与肝癌发病过程中的关系。结果与正常肝组织及肝硬化组织相比,Bmi-1基因在肝癌组织中呈明显高表达。此外,过表达的Bmi-1与肝癌患者的临床分期(P<0.001)、肿瘤大小(P=0.03)及淋巴结转移(P=0.021)明显相关。结论过表达的Bmi-1基因mRNA可能参与了肝癌的发病过程,并与肝癌的进展及淋巴结转移相关。
【Objective】 To investigate the role of Bmi-1 in the pathogenesis of liver cancer.【Methods】 Total RNA of fresh liver cancer samples,liver cirrhosis samples,and liver tissues were extracted by Trizol.After total RNA was transcripted into cDNA,the expression of Bmi-1 gene was detected in these samples by real-time PCR.Finally,the correlation of Bmi-1 expression and clinical features of liver cancer patients was analyzed.【Results】 Comparing to the liver cirrhosis samples and liver tissues,the expression of Bmi-1 was markedly increased in liver cancer samples(P 0.05).In addition,Bmi-1 expression in liver cancer samples was obviously related to the clinical stage(TNM,P 0.001) T classification(Tumor size,P =0.03),lymph node involvement(N classification,P =0.021).【Conclusions】 Overexpressed Bmi-1 may be involved in the pathogenesis of liver cancer and promoted the progression and the metastasis of liver cancer.
