葛根素对糖尿病大鼠心肌纤维化的影响 被引量:7

摘要 目的:观察葛根素对糖尿病大鼠心肌损伤的保护作用,探讨心肌纤维化可能的发病机制。方法:制造糖尿病大鼠心肌病模型并分别给予不同剂量的葛根素灌胃,12周后检测血糖、心脏质量指数变化,光镜下观察心肌细胞形态学及胶原纤维的改变,免疫组化法和蛋白质印迹法测定心肌组织中基质金属蛋白酶2(MMP-2)和基质金属蛋白酶9(MMP-9)蛋白含量的变化。结果:糖尿病组大鼠血糖、心脏质量指数显著升高(P<0.05),心肌胶原纤维显著增多(P<0.05),MMP-9蛋白含量增高明显(P<0.05),MMP-2蛋白含量降低明显(P<0.05);葛根素各治疗组与糖尿病组相比,血糖、心脏质量指数有不同程度降低(P<0.05),心肌胶原纤维含量减少(P<0.05),MMP-9蛋白含量减少(P<0.05),MMP-2蛋白含量增高(P<0.05),尤以高剂量组改变最为明显。结论:心肌组织中的MMP-2、MMP-9的表达改变与糖尿病心肌纤维化有关,葛根素通过影响上述指标的变化对糖尿病心肌病产生治疗作用。 Objective: To observe the protective effects of puerarin on cardiac muscle in diabetic rats and explore the mechanism of myocardial fibrosis.Methods: Diabetic model rats were established by intra-abdominal injection of streptozotocin.Puerarin-treated rats were administered by gastric canal at different dose of puerarin respectively.After 12 weeks,blood sugar and heart weight index were examined in all groups.The structure of heart and the change of collagen fiber were examined by light microscope.Semiquantitative expressions of MMP-2 and MMP-9 protein were determined by immunohistochemistry and Western Blot.Results: Fasting blood glucose,heart weight index of the diabetic rats were significantly up-regulated(P0.05),collagen fiber were much higher(P0.05),the protein expression of MMP-9 were significantly up-regulated(P0.05),MMP-2 were significantly decreased.Compared with the diabetic group,fasting blood glucose and heart weight index of puerarin therapy groups were significantly decreased(P0.05),collagen fiber were declined significantly(P0.05),the protein expression of MMP-9 were decreased and MMP-2 were up-regulated(P0.05).Conclusion: The expression of MMP-2 and MMP-9 on cardiac muscle are related to myocardial fibrosis of diabetic,puerarin has therapeutic effect on diabetic cardiomyopathy through affecting the changes of above mentioned indexes.
