摘要
使用电喷雾质谱检测了糖用甜菜中甜菜碱,并且采用串联质谱技术分析了甜菜碱的碎裂规律。实验结果表明:甜菜碱在酸性条件下检测,在质谱中表现为[M+H]+离子,谱图比较简单,然而在中性条件下检测,簇和及加钠和加钾离子现象严重,谱图比较复杂,因此确定了甜菜碱的检测条件为酸性。在串联质谱中甜菜碱碎裂产生含氮和不含氮的两种特定的子离子,可以作为甜菜碱的定性依据。甜菜提取液不需经过复杂的前处理就可以检测出甜菜碱,操作简单、快速,灵敏度高、特异性好,可以作为复杂体系中甜菜碱的快速检测方法。
Betaine is detected by using electrospray ionization mass spectrometry(ESI-MS) and tandem mass spectrometry(ESI-MSn).The analysis condition is optimized and its fragment mechanism also is concluded.The mass spectrum of betaine is very simple in acid solution,e.g.,only + peak is predominant.Therefore,there are more peaks of cluster ion and + and + in neutral solution,and the mass spectrum is rather complex,that is adverse to analysis.Thus,it is concluded that the optimum condition for detecting betaine is acid solution in ESI positive ion mode.In ESI-MSn,protoned betaine ion is collided to produce 2 distinct fragments,which is especially helpful to quality betaine.ESI-MS can be used to directly analysis sugarbeet extraction with high sensitivity and good reproducibility,and avoiding tedious and time-consuming chemical manipulations.
