摘要
目的建立兔肝硬化动物实验模型,在此基础上建立恶性肿瘤肝转移模型,探讨肝硬化对恶性肿瘤肝转移过程中的影响及其机制。方法采用CCL4皮下注射法建立兔肝硬化模型,随机分为实验组和对照组成,实验组用CCL4溶液皮下注射,对照组给予等量橄榄油。每3周采集兔血和小块肝组织,进行生化指标检测和肝脏HE染色病理学观察。明确肝硬化形成后,分别通过经胃壁、脾脏接种VX2肿瘤细胞,观察肝硬化兔与正常肝脏兔接种VX2肿瘤细胞后肝转移发生率及肿瘤生长情况。结果 (1)实验9周末HE染色可见肝硬化典型病理变化假小叶形成,肝硬化模型建立成功。(2)肝硬化组恶性肿瘤肝转移率明显低于对照组(P<0.05)。结论 CCL4溶液皮下注射成功诱导建立了兔肝硬化模型,并在此基础上建立了恶性肿瘤肝转移模型,实验结果提示肝硬化对恶性肿瘤的肝转移具有一定的抑制作用。
Objective To establish the model of hepatic cirrhosis and then build a malignant tumor liver metastases model on the basis of the former model.To investigate the impact mechanism of hepatic cirrhosis in malignant neoplasm hepatic metastasis.Methods Establish the rabbit model of liver cirrhosis induced by injecting carbon tetrachloride CCL4 into rabbits.New Zealand rabbits in the experimental group were received subcutaneous injection of CCL4 in nine weeks.Rabbits in the control group were injected subcutaneously with the same quantity of olive oil.Blood samples were collected to examine the blood protein and enzymes once in 3 weeks.The development of liver fibrosis was recorded by the HE method once 3 in weeks.After being identified the formation of hepatic cirrhosis,the liver metastases model was developed by transplant the VX2 neoplasm cell into the serous membrane of the stomach or spleen.Hepatic metastasis incidence and tumor growth conditions both in hepatic cirrhosis rabbits and in normal rabbits that were vaccinated with VX2 neoplasm cell were observed.Results(1) At the end of 9 weeks,typical features of liver cirrhosis and the formation of false lobe were observed.(2) The hepatic metastasis incidence in hepatic cirrhosis group was obviously lower than that in the control group.Conclusion The experiments establish rabbit hepatic cirrhosis successfully by using subcutaneous injection of CCL4 and establish a neoplasms hepatic metastasis model on the basis of the method.And it proofs that hepatic cirrhosis has something inhibition effect on hepatic metastasis in some extent.
