新疆天山乌头草的急性毒性试验研究 被引量:6

Study on acute toxicity test of Xinjiang Tianshan Mountains' Aconitum carmichaelii herb

摘要 目的:弄清新鲜乌头草对小鼠的急性毒性。方法:野外采集新鲜乌头草,立即粉碎榨汁,得到新鲜乌头草汁,小鼠灌胃给药,测定新鲜乌头草的半致死量LD50,观察小鼠所出现的毒性反应,并测定小鼠血清的生化指标和心脏的氧化还原指标的变化。结果:以新鲜乌头草量计,乌头草对小鼠的LD50是7.550g/kg;乌头草高剂量组小鼠血清中丙氨酸氨基转移酶(ALT)、门冬氨酸转移酶(AST)、总蛋白(TP)、白蛋白(ALB)、尿素(Urea)、尿酸(Uric Acid)、肌酸激酶(CK)、α-羟丁酸脱氢酶(α-HBDH)、乳酸脱氢酶(LDH)、肌酸激酶同工酶(CK-MB)与对照组相比均有显著性升高;与对照组相比,心脏组织中丙二醛(MDA)含量显著升高、总抗氧化能力(T-AOC)显著降低,以及乳酸脱氢酶(LDH)活力显著降低。结论:新鲜乌头草有很大的急性毒性,主要毒性靶器官是心脏组织。 Objective:To explore the acute toxicity of fresh Caulis et Folium Aconitum carmichaeli Debx.in mice.Methods:The fresh Caulis et Folium Aconitum carmichaeli Debx.was shivered and pressed.The obtained juice were treated with a single oral dose,The medial lethal dose(LD50) of fresh Caulis et Folium Aconitum carmichaelii was determined in mice;the toxic reaction was observed;the serum samples were collected for biochemical analysis and the cardiac tissues were used to quantitated the redox state.Results:The LD50 of fresh Caulis et Folium Aconitum carmichaeli Debx was 7.550g/kg in mice.Compare to the control group,the activity of ALT,AST,CK,α-HBDH,LDH,CK-MB and the content of TP,ALB,Urea,Uric Acid were increased with significant difference in the mice serum of high dose fresh Caulis et Folium Aconitum carmichaeli Debx.group.The content of MDA increased,T-AOC decreased,and the vigor of LDH decreased significantly as compare to the control group.Conclusions:In general the fresh Caulis et Folium Aconitum carmichaeli Debx.could be considered to have acute toxicity,and the main target organ is cardiac tissue.
