摘要
目的探讨糖足洗液对链脲佐菌素(STZ)诱导的糖尿病小鼠皮肤溃疡的疗效及机理。方法昆明种小鼠50只,随机抽取10只为正常对照组,其余小鼠在禁食12h后均按150mg/kg腹腔注射STZ造模,并随机分为模型对照组、单用胰岛素组、糖足洗液组、金黄散对照组,每组10只。5组小鼠均在背部单侧制造直径约8mm的皮肤溃疡模型。除正常对照组和模型对照组外,其余3组均注射胰岛素,其中2组分别予糖足洗液及金黄散湿敷。动态观测各组小鼠体重、创面大小、空腹血糖(FBG)等指标。连续用药30d后禁食8h,测定C反应蛋白(CRP)、白介素6(IL-6)和肿瘤坏死因子-α(TNF-α)等指标。结果糖足洗液组糖尿病小鼠溃疡时间早于模型对照组,差异有统计学意义(P<0.01),FBG、CRP、IL-6、TNF-α均低于模型对照组,体重高于模型对照组,差异均有统计学意义(P<0.05或P<0.01)。结论在胰岛素有效控制血糖的前提下,应用糖足洗液能够缩短溃疡愈合时间、促进肉芽组织生长、促进创面愈合,其机制与降低炎症细胞因子水平有关。
Objective To study the curative effect and mechanism of diabetic-foot lotion on the skin ulcer of mice with diabetic foot(DF) induced by streptozotocin(STZ).Methods Ten of the fifty healthy male Kunming mice were put into the normal group and the other 40 were injected STZ 150mg/kg intraperitoneally after 12h fasting to make model,and were randomized into 4 groups,model control group,insulin group,Diabetic-foot Lotion group,and Jinhuang San control group,with 10 in each.All mice were made skin ulcer models by cutting on one side of the back to produce a skin ulcer approximately 8mm in diameter.Except for the normal control group and model control group,the other three groups were injected insulin,of them,two groups were given Diabetic-foot Lotion and Jinhuang San for exteral application.The body weight,ulcer size,and fasting blood glucose(FBG).were dynamically observed.After treatment for 30 days in succession and fasted for 8h,c-reactive protein(CRP),interleukin(IL-6),and tumor necrosis factor α(TNF-α) were determined.Results The ulcer healing of the Diabetic-foot Lotion group was earlier than that of the model control group,the difference was significant(P〈0.01).and the level of FBG,CRP,IL-6,and TNF-α was lower than that of the model control group,the body weight being higher than that of the model control group with significant differences(P〈0.05 or P〈0.01).Conclusion Under the premise of effective control of blood glucose with insulin,the application of Diabetic-foot Lotion quickens the ulcer healing through promoting the granulation tissue growth.Its mechanism is related to the decreases of the inflammatory cell factors.
