基质辅助激光解析电离时间飞行质谱技术检测miR146a单核苷酸多态性

摘要 目的:运用基质辅助激光解析电离时间飞行质谱技术(MALDI-TOF-MS)检测miR146a基因单核苷酸多态性,并探讨其与哈萨克族食管癌的相关性。方法:运用MALDI-TOF-MS检测261例哈族食管癌患者及190例同地区对照样本miR146a基因rs2910164位点单核苷酸多态性。结果:成功应用MALDI-TOF-MS检测了哈族食管癌miR146a基因rs2910164位点G/C多态,其基因型分布频率分别是36.4%,49.0%和14.6%,与对照组基因型分布频率(分别是36.3%,49.5%,14.2%)之间差异无统计学意义(P>0.05)。结论:基于MALDI-TOF-MS技术检测miR146a基因多态性的方法是一个快速、准确和可靠的检测方法。 Objective:Using the matrix-assisted laser desorption ionization time-of-flight mass spectrometry(MALDI-TOF MS) analysis to detect the single nucleotide polymorphisms of miR146a,and explorer the relationship between Hazakh's esophageal carcinoma and the single nucleotide polymorphisms of miR146a gene. Methods:Using MALDI-TOF MS to detect the SNP of 261 cases of Hazakh's esophageal squamous cell carcinomas(ESCC),190 cases of cancer-free controls in the same area. Results:MALDI-TOF MS detected the SNP of miR146a sucessfully,miR146a gene rs2910164 G/C polymorphisms genotype frequencies were 36.4%,49.0% and 14.6%,the different has no statistic significant compare with the genes of control(36.3%,49.5%,14.2%). Conclusion:the method based on MALDI-TOF-MS technique for detecting SNP miR146a gene is rapid,accurate and reliable method.
