摘要
目的观察白藜芦醇(Res)对博莱霉素(BLM)致大鼠肺纤维化和NF-κBmRNA表达水平的影响。方法 90只雌性SD大鼠随机分为对照组、模型组、Res低(25 mg/kg)、中(50 mg/kg)、高(100 mg/kg)剂量干预组及地塞米松(DM 3 mg/kg)干预组,以气管内注入BLM法制备肺纤维化大鼠模型,采用Masson染色观察各组肺组织的纤维化改变情况,并用RT-PCR方法测定肺组织中NF-κB mRNA的表达水平。结果 1.模型组及Res低剂量组肺组织胶原纤维随时间的推移明显增多,Res高、中剂量组及DM干预组肺组织病理变化较模型及Res低剂量组明显减轻;2.模型组及Res低剂量组NF-κB表达水平明显高于对照组,并呈现逐渐增高的趋势,Res高、中剂量组及DM干预组变化趋势与模型组类似,但表达水平均低于同时间点的模型组及Res低剂量组。结论 NF-κB的高表达在肺纤维化的发病机制中起重要作用,Res抑制NF-κB的高表达可能是其防治肺纤维化的机制之一。
Objective To observe the effect of resveratrol on bleomycin-induced pulmonary fibrosis of the rat and the expression of NF-κBmRNA.Methods 60 female SD rats were randomly divided into control group,model group,and low-(25 mg/kg),middle-(50 mg/kg),high-(100 mg/kg) resveratrol-treated group,dexamethasone(3 mg/kg)-treated group.Rat pulmonary fibrosis was reproduced by an intratracheal injection of bleomycin.Using the method of Masson to observe the fibrosis in lung tissue changing,and RT-PCR was used to detect the expression of NF-κB mRNA in lung tissue.Results 1.The collagen fibers in the model and low resveratrol-treated group was significantly increased as time progressed,the middle and high resveratrol-treated group and the dexamethasone-treated group have the same trends with the model group and the low resveratrol-treated,less than them at the same time.2.The expression of NF-κB mRNA in the model and low resveratrol-treated group was more than the control group,and showed the trend of gradually increasing,the middle and high resveratrol-treated group and the dexamethasone-treated group have the same trends with the model group and the low resveratrol-treated,but less than them at the same time.Conclusion High expression of NF-κB plays an important role in the pathogenesis of pulmonary fibrosis.Resveratrol can inhibit the expression of NF-κB.Maybe it's one of the mechanisms that resveratrol prevents and treats lung fibrosis.
