摘要
目的探讨基质金属蛋白酶-2(matrix metalloproteinase-2,MMP-2)和色素上皮衍生因子(pigment epithelium derived factor,PEDF)在氧诱导小鼠视网膜新生血管中的表达和意义。方法随机选取80只健康C57BL/6J小鼠进行分组,取对照组(40只)和高氧组(40只)小鼠眼球作ADP酶视网膜铺片、病理切片及免疫组织化学法检测,观察视网膜血管的改变,计算视网膜新生血管突破内界膜的内皮细胞核数及检测MMP-2、PEDF蛋白的表达。结果高氧组视网膜大量新生血管形成;新生血管突破内界膜的内皮细胞核数为(32.45±1.34)个,与对照组(1.27±0.20)个相比差异有统计学意义(t=10.345,P<0.05)。高氧组与对照组相比,MMP-2蛋白在神经节细胞层、内丛状层、内核层和突破视网膜内界膜的新生血管中高表达(P<0.05);PEDF蛋白在神经纤维层、神经节细胞层、内丛状层、感光细胞层及视网膜色素上皮细胞层低表达(P<0.05)。两者表达呈显著负相关(r=-0.785,P<0.05)。结论 MMP-2和PEDF共同维持视网膜新生血管的形成。
Objective To explore the expression and significance of matrix metalloproteinase-2(MMP-2) and pigment epithelium derived factor(PEDF) in oxygen-induced retinal neovascularization of mice.Methods Eighty healthy C57BL/6J mice were chosen and randomly divided into control group and hyperoxic group,40 mice in each group.The eyeballs of two groups were enucleated to make sample.Adenosine diphosphate-ase(ADPase) stained retina flat-mounts was performed to assess the retinal vascular profiles,pathological section was applied to count the number of new vascular cell nuclei breaking through the inner limiting membrane and the expression of MMP-2 and PEDF was detected by immunohistochemical method.Results Numerous neovascularization was seen in the hyperoxic group.The number of nucleus of new vessels vascular endothelial cells breaking through the inner limiting membrane in the hyperoxic group was 32.45±1.34,which was significant higher than that in control group(1.27±0.20),there was statistical difference(t=10.345,P0.05).Compared with the control group,there were high protein expression of MMP-2 in the ganglion cell layer,inner plexiform layer,inner nuclear layer and neovascularization breaking through the inner limiting membrane in the hyperoxic group,there was statistical difference(P0.05);PEDF protein were faintly expressed in the nerve fiber layer,ganglion cell layer,inner plexiform layer,photoreceptor cells layer and retinal pigment epithelial layer in the hyperoxic group,there was statistical difference(P0.05).And there was an obvious negative correlation between the expression of MMP-2 and PEDF(r=-0.785,P0.05).
