银染PCR-SSCP法检测食管癌P53基因突变

摘要 目的 :探索快速、有效、简便的检测食管癌 P5 3基因突变的方法 ,以利于食管癌的早期诊断和研究。方法 :用银染 PCR- SSCP法检测本院 63例食管切除标本的 P5 3基因突变 (选突变多发位点 5、6、7、8外显子 )。结果 :食管癌 P5 3基因突变发生于第 5、6、7、8外显子 ,总突变率 68% ,鳞癌、腺癌、未分化癌中 P5 3突变率差异不显著 ;鳞状上皮不典型增生中 P5 3突变率为 61.5 %。结论 :P5 Objective:To establish a simple and efficient method to detect the expression of P53 gene mutation in esophageal carcinoma for its early diagnosis.Methods:Single strand conformation polymorphism(PCR SSCP)combined with silver staining as a method was established to screen the expression of P53 gene mutation(the exons 5,6,7,8)in 63 esophageal carcinomas.Results:The expression of P53 gene mutation in esophageal carcinomas frequently happened at the exons 5,6,7,8,mutation rate 68%(34 of 50).There is no significant difference among squamous cell carcinoma,adenocarcinoma and undifferentiated carcinoma.In addition,P53 gene mutation rate is 61.5%(8 of 13)in the atypical hyperplasia of squamous epithelium.Conclusion:The expression of P53 gene mutation may be used as a valuable index of molecular biology;PCR SSCP argentation is a reasonable method for the early diagnosis of esophageal carcinoma.
