不同生长期的灵芝三萜含量测定及变化规律研究 被引量:12

摘要 本试验研究了不同生长期的灵芝三萜含量变化规律。流动相为乙腈-2%的冰醋酸(梯度洗脱);检测波长为252 nm;流速为1 mL/min;柱温为35℃。高效液相指纹图谱研究表明紫芝亦含有三萜酸成分,但三萜酸含量明显偏低,且紫芝的主要特征单位生药峰面积只有赤芝的1/10~1/20左右。不同入土时间的赤芝三萜指纹图谱差异明显,部分三萜酸特征峰以入土30 d的峰面积为最大,部分特征峰以入土50~60 d的峰面积为最大。紫芝则是入土60~70 d时其大部分三萜酸特征峰面积最大。总三萜含量测定结果表明赤芝在入土30 d与入土50 d时总三萜含量较高,70 d时含量最低。紫芝在入土70 d时总三萜含量最高,50 d后增加趋于平缓。总三萜含量测定与三萜指纹图谱结果类似,研究结果为灵芝的采栽期提供了一定参考,提示对于灵芝的质量评价应采用指纹图谱与含量测定相结合为宜。 The content and variation of triterpenoids of Glossy ganaderma harvested at different growth periods were studied by HPLC using acetonitrile-2% solution(gradient elution) as a mobile phase at 252 nm with flow rate and column temperature being of 1mL/min and 35 ℃,respectively.HPLC fingerprint spectrum research indicated Ganoderma sinense also contained triterpenic acid,but its content was significant lower than that in Ganoderma lucidum,and its main characteristic peak area per unit medicine was only five to ten percent of Ganoderma lucidum.Ganoderma lucidum harvested at different growth periods.Their HPLC fingerprint spectrums of triterpenic acids were significant different.The character peak area of some parts of triterpenic acids was the largest in Ganoderma lucidum after cultivated for 30 days,while the others were the biggest after cultivated for 50 to 60 days.Most triterpenic acid peak areas were the biggest in Ganoderma sinense after cultivated for 60 to 70 days.The content of total triterpenoids of Ganoderma lucidum was bigger after cultivated for 30 days and 50 days,and the least in the 70th day.The content was the biggest in Ganoderma sinense after cultivated for 70 days,but it changed little after 50 days.The result of total triterpenoids content determination was very similar with that of HPLC fingerprint spectrum which provides references of picking period choosing for Glossy ganoderma,also indicates that quality evaluation of Glossy ganoderma should combine HPLC fingerprint spectrum with content determination method.
