杂色云芝菌产漆酶条件优化及其对碱性紫5BN的降解 被引量:1

摘要 通过实验对杂色云芝菌产漆酶的发酵条件进行了优化,结果表明杂色云芝菌产漆酶的最佳发酵条件为:以质量浓度为15 g/L的羧甲基纤维素钠和质量浓度为1.25 g/L的葡萄糖作为复合碳源,以质量浓度为15 g/L的酵母膏作为氮源,在发酵72 h时加入浓度为2 mmol/L的2,5-二甲基苯胺,发酵96 h时加入浓度为2 mmol/L的Cu2+。发酵培养168 h后酶活力达到3 319.2 U/mL。采用该菌株产漆酶对碱性紫5BN溶液进行脱色处理,当漆酶粗酶液酶活力为0.01 U/mL时,反应10 h,脱色率可达93.91%。 The fermentation conditions for laccase production from Coriolus Versicolor were optimized by experiments. The optimum fermentation conditions are as follows: Using sodium carboxymethy cellulose with 15 g/L of mass concentration and glucose with 1. 25 g/L of mass concentration as the complex carbon source; using yeast extract with 15 g/L of mass concentration as the nitrogen source; adding 2,5-dimethylaniline with 2 mmol/L of concentration at the fermentation time for 72 h; adding Cu2+ with 2 mmol/L of concentration at the fermentation time for 96 h. When the fermentation time is 168 h, the enzyme activity can reach 3 319.2 U/mL. The solution containing basic violet 5BN was treated using the laccase produced by this strain. When the enzyme activity of the crude enzyme solution is 0. 01 U/mL and the treatment time is 10 h, the decolorization rate is 93.91%.
