针刺对实验性大鼠脑出血后灶周组织NF-κBp65表达和脑水含量的影响及其相关性分析 被引量:6

摘要 目的:探讨针刺对急性脑出血(Intracerebral hemorrage,ICH)后核转录因子-κB(NF-κBp65)动态表达及脑水含量变化的影响,并探讨两者的相关性。方法:实验选用Wistar健康雄性大鼠96只,随机将其中90只分为模型组、针刺组、西药组(茴拉西坦组)。每组又分为6h、1天、2天、3天和7天5个时间点,每个时间点随机分配6只大鼠,另设6只大鼠做空白对照。制备ICH大鼠模型,采用蛋白免疫印迹技术观察针刺在不同时间点对ICH大鼠脑组织中NF-κBp65蛋白表达及脑水含量的影响,并对两者做相关性分析。结果:针刺组大鼠脑内NF-κBp65蛋白表达在不同时间点均有显著下调,与模型组比较有显著性差异(P<0.05),相关性分析提示,脑水肿与NF-κBp65表达呈正相关。结论:ICH后NF-κBp65阳性表达是加重脑水肿的一个重要因素,针刺可下调ICH后血肿周围脑组织内NF-κBp65蛋白表达,减轻脑水肿程度,防治ICH后继发性脑损伤。 Objective: To study the dynamic expression of NF-κBp65 and the relation between the brain water contents and NF-κBp65 after intracerebral hemorrhage(ICH) rats and intervention effect of acupuncture.Method: The 96 healthy Wistar male rats were selected.90 healthy male Wistar rats were randomly divided into 3 groups : model group,acupuncture group and western medicine group,30 rats in each group.Each group was divided into 5 time phase point of 6 hours,1day,2days,3days and 7days,respectively,6 rats in each group.Meanwhile,6 rats were taken as the blank control group.The intracerbral hematoma rat model was made by autologous blood injection by using stereotactic method Western-blot analysis to observe the expression of NF-κBp65 of the intracerebral hemorrage rats.Result: The expression of NF-κBp65 had an obvious down-regulation at different phases in the brain of the acupuncture group rats,and it showed significant differences compared with model group(P0.05).The positive correlation between NF-κBp65 and brain water content was observed(P0.01).Conclusion: The acupuncture of BaiHui-QuBin can antagonize brain edema and secondary brain injury caused by inflammatory response and adhesive response after ICH by means of inhibiting the influence of NF-κBp65 during the acute period of intracerebral hemorrhage.
