新疆发酵乳酪乳清体外ACE抑制和抗氧化活性研究 被引量:3

Study on ACE inhibition and antioxidant activities in vitro of traditional fermented cheese whey in Xinjiang

摘要 目的:研究新疆传统发酵乳酪乳清(简称TFCW)超滤提取物对血管紧张素转换酶(ACE)的抑制活性、抗氧化活性。方法:以体外活性ACE抑制率和DPPH.,.OH,O2-.的清除率为指标,检测TFCW中相对分子质量范围分别在30~10 kDa,10~5 kDa和5 kDa以下超滤提取物的ACE抑制活性、抗氧化活性。用Sephadex G-15柱层析对5 kDa以下肽段进行分离,并对其分离产物进行检测。结果:相对分子质量5 kDa以下的肽段,ACE抑制作用和抗氧化活性最高。用Sephadex G-15对其进行分离,得到5个组分。其中组分4(相对分子质量分布在1071~724 Da)对ACE抑制率高达56.45%;组分5(相对分子质量分布在645~500Da)对自由基DPPH.,.OH,O2-.的清除率分别是59.06%,62.06%,29.51%。结论:TFCW中的多肽具有较强的体外ACE抑制活性和抗氧化活性。 Objective :To study the ACE inhibitory activity and antioxidant activity of the ultrafiltration extracts of traditional fermented cheese whey (TFCW) in Xinjiang. Methods: ACE inhibitory ratio and DPPH·, · OH, O2^- clearance in vitro were used as indicatorsto determine the activity of the extracts with molecular weight of 30 -10 kDa,30 -10 kDa and less than 5 kDa. Sephadex G-15 column was used to separate the uhrafiltration extracts whose molecular weights were less than 5 kDa. The activities of the separation products were determined. Resuits:The peptides with molecular weight below 5 kDa had the highest ACE inhibition and antioxidant activity. These peptides were separated into 5 components by Sephadex G-15 column. Component 4 (molecular weight dis- tribution in 1 071 -724 Da) had a high rate of ACE inhibition,accounting for 56.45% of 5kDa peptides. Compo- nent 5 (molecular weight distribution in 645 -500 Da) had a high free radical scavenging capacity,with DPPH· , ·OH and O^- clearance of 59.06% , 62.06% , and 29.51% , respectively. Conclusion:The peptides in TFCW have strong ACE inhibitory activity and antioxidant activity in vitro.
