摘要
目的 探讨医用硅橡胶体外致突变试验的方法。方法 选用体外姐妹染色单体互换试验评价医用硅橡胶材料的诱变性。从细胞遗传学角度,对材料的浸提介质及其与细胞的接触剂量进行实验性探讨。结果 以生理盐水作为浸提介质时,加入剂量以1ml为适;用Eagle'sMEM作为浸提介质时,由于可直接调控材料与细胞的接触剂量,具有较大的实用性。结论 体外姐妹染色单体互换试验可推荐作为一项检测口腔材料潜在致突变性的体外评价方法。
Objective To investigate the method of in vitro mutagenicity test for medical silicone. Method The mutagenicity of medical silicone by using sister chromatid exchange test was evaluated. From the cytogenetoxicity point of view, the extracted medium and contacted concentration between the material and cells were discussed. Results The contacted concentration could be adjusted directly by using Eagle's MEM as an extracted medium, while only 1ml of saline as extracted medium was suitable for this test. Conclusion It is recommended that sister chromatid exchange test is an effective in vitro detecting method for mutagenicity of dental materials.
