摘要 目的研究橙皮苷(HDN)自微乳在大鼠小肠各部位的吸收情况及影响因素。方法运用大鼠在体肠循环模型,以HPLC法测定循环液中的酚红和药物浓度,观察不同肠段、给药浓度及不同剂型因素对药物小肠吸收的影响。结果 HDN自微乳在十二指肠、空肠、回肠、结肠部位均有吸收,其药物吸收速率常数(Ka)差异无统计学意义;给药浓度对药物小肠吸收的Ka值随浓度增大而减小,但差异无统计学意义;与HDN混悬剂相比,HDN自微乳的Ka值和肠吸收百分率均显著增加,差异有统计学意义。结论 HDN自微乳在小肠各部位有吸收,且以被动扩散方式为主,自微乳能显著提高HDN的吸收。 Objective To study the absorption kinetics of hesperidin(HDN) self-microemulsion in rat′s intestine.Methods The tests were carried out in situ rays intestine technique.The effects on absorption of different rat′s intestine segments,concentration of HDN and preparation factors were studied.Results There were no significant differences among the absorption constants(Ka) of HDN self-microemulsion in the duodenum,jejunum,ileum and colon of rats.The Ka of HDN self-microemulsion in ileum was no significantly influenced by different concentration of HDN;Compared with HDN powder,Ka and intestinal absorption of HDN self-microemulsion were significantly improved.Conclusion The absorption of HDN self-microemulsion in rat′s intestine is mainly of the passive diffusion mechanism without specific absorption site.Self-microemulsifying drug delivery system can significantly improve the absorption of HDN.
