松果腺摘除联合阿霉素肾病动物模型的建立 被引量:3

摘要 目的研制出一种特殊的大鼠松果腺摘除联合阿霉素(ADR)肾病模型。方法在手术摘除松果腺的基础上注射ADR,检测0,1,2,4wk时大鼠血生化和各周时24h尿蛋白水平。结果ADR组和ADR+松果腺摘除组大鼠均呈明显肾病综合症表现,且两者相比,后者的24h尿蛋白排泄量和血浆胆固醇水平显著高于前者,而血浆总蛋白和白蛋白水平则与之相反;单纯松果腺摘除不影响大鼠尿蛋白水平;松果腺摘除手术相关并发症是导致大鼠死亡的主要原因。结论松果腺摘除可加重阿霉素肾病的肾损害;这种特殊类型的肾病模型是研究松果腺褪黑素与慢性肾病关系的又一工具。 AIM To produce a special animal model of adriamycin induced nephropathy after pinealectomy in Wistar rats. METHODS Before the rats were injected adriamycin (5 mg·kg -1 ) vialtail vein, their pineal glands were removed by surgical operation. At 0,1,2 and 4 week, urinary excretion protein (UAE) and plasma parameter in all rats were investigated. RESULTS The typical nephrotic syndrome was found in group adriamycin administration(ADR) and in group pinealectomy with adriamycin administration (PX+ADR). UAE level in group PX+ADR increased more significantly ( P <0 01) than that in group ADR (1024 97±137 02, 407 07±68 66 mg·d -1 , respectively). The similar change was found in plasma cholesterol of group ADR and PX+ADR. Plasma total protein and albumin levels in group ADR decreased more markedly ( P <0 01) than that in group PX+ADR. The main dead cause of rats was operation complications with pinealectomy. CONCLUSION This special animal model is a great tool to study relationship between pineal gland and chronic nephropathy
