摘要
目的:探讨高危型HPV16 E6蛋白的表达对宫颈癌细胞株中p63α表达的影响。方法:用RT-PCR法从SiHa中扩增出带酶切位点的HPV16 E6的片段,将HPV16 E6基因片段克隆到p3×flag-CMV-Myc-24真核表达载体上,通过脂质体转染入宫颈癌细胞株后,W estern blot检测p63α的表达。结果:成功构建HPV16 E6真核表达载体,转染至宫颈癌SiHa和Caski细胞株中,p63α均低于对照组。结论:高危型HPV E6能够抑制p63α的表达,提示p63α与HPV E6的相互作用在子宫颈癌发病中发挥重要作用。
Objective:To explore the effects of expression of high risk HPV E6 protein on the expression of p63α in the cervical cancer cell lines.Methods:HPV16 E6 gene sequence were amplified from total RNA of SiHa cell lines by reverse transcription-polymerase chain reaction(RT-PCR),then the HPV16 E6 gene sequence was inserted into the eukaryotic expression vector p3×flag-CMV-Myc-24.Western blot were used to test the expression levels of p63α after transfected into cervical cancer cell lines.Results:The HPV 16 E6 eukaryotic expression vector was successfully constructed,low expression of p63α after transfection in cervical cancer cell lines SiHa and CaSki copare to the control group.Conclusion:HR E6 protein can inhibits cervical cancer cell lines expression of p63α,p63α and HPV E6 interactions may play an important role in the pathogenesis of cervical cancer.
