摘要
目的观察缺氧对体外血脑屏障(blood brain barrier,BBB)紧密连接蛋白ZO-1表达的影响以及清开灵的干预作用。方法通过体外培养Balb/c小鼠脑微血管内皮细胞系形成BBB,采用缺氧缺糖24 h建立BBB缺血损伤模型,分别以荧光实时定量和Western blot的方法测定BBB紧密连接蛋白ZO-1的基因转录和蛋白表达水平,同时观察不同剂量清开灵对ZO-1表达的影响。结果与正常组相比,缺氧缺糖24 h可使BBB表达ZO-1明显减少(P<0.01)。清开灵能够显著增加缺血后ZO-1的基因转录和蛋白表达水平,与模型组差异显著(P<0.05或P<0.01)。结论清开灵可通过改善脑缺血对BBB主要组成蛋白的表达,发挥其对缺血性脑中风的脑保护作用。
Objective To observe the effects of hypoxia on expression of tight junction ZO-1 of blood brain barrier(BBB) in vitro and the role of Qingkailing injection.Methods The BBB in vitro was established by culturing mouse cerebral microvasculature endothelial cells of Balc.The model of ischemic injury was established by oxygen and sugar be deprived for 24 h.The gene transcription and protein expression level of ZO-1 were measured by fluorescence real-time quantitative and western blot respectively.Meanwhile,the expression of ZO-1 caused by different dose of Qingkailing injection was detected.Results compared with control group,the expression of ZO-1 in BBB was reduced significantly after oxygen and sugar be deprived for 24 h(P0.01).Compared with model group,the expression of gene transcription and protein expression of ZO-1 were increased in Qingkailing injection group,and there was statistical difference(P0.05 or P0.01).Conclusion Qingkailing injection had cerebral protection role in ischemic stroke by improving the expression of main component protein of BBB damaged by brain ischemia.
