摘要
为了优化生产工艺,提高药液的澄清度,更好的保留有效成分,分别采用天然高分子絮凝剂ZTC和壳聚糖对养阴清肺糖浆原药提取液进行絮凝处理。以上清液的浊度和有效成分的含量作为衡量指标,考察了絮凝剂用量、絮凝温度和搅拌条件对絮凝效果的影响,并将絮凝后的上清液离心分离,最终确定了最佳工艺条件。结果发现絮凝-离心分离法处理的养阴清肺糖浆原药提取液中甘草酸和芍药苷的保留率分别为83%和95%,浊度为1.3 NTU。与醇沉法相比,絮凝后水提液中甘草酸的含量提高了25%,放置一个月后浊度仅为醇沉法的一半。絮凝-离心分离法用于养阴清肺糖浆原药提取液的净化效果明显,有效成分含量、药液澄清度及稳定性较高。
The natural macromolecule flocculent ZTC1+1 and chitosan were applied to purify the herbs extract solution of Yang-yin-qing-fei syrup so as to improve the herbs extract clarity and reserve more active components of the solution.By analyzing the turbidity degree and the active ingredients content of the supernatant solution,the influences of the flocculant dose,the solution temperature,and the agitating condition on the flocculation indexes were investigated and the optimum process conditions were obtained.The results show that the retention rate of glycyrrhizic acid and pareonia radix in the supernatant solution kept at 83% and 95% respectively,and the turbidity could reduce to 1.3NTU under the optimum conditions of flocculation-centrifugation method.Compared with the ethanol precipitation of the herbs extract solution,the content of glycyrrhizic acid increased by 25%,and a month later the turbidity was only half of it by ethanol precipitation.Flocculation-centrifugation technology has distinct advantages of high content of active components,high clarification degree and long solution stability for the herbs extract solution purification of the Yang-yin-qing-fei syrup.
