摘要
以水稻幼苗为材料,研究内源茉莉酸的超高效液相色谱-质谱/质谱(UPLC-MS/MS)定量分析。用甲醇浸没植物组织样品提取JA,提取液经过固相萃取小柱(SPE-NH2)去除杂质,二氢茉莉酸(H2JA)作为内标物,通过UPLC-MS/MS仪进行定量分析。设置5,20,100 ng/g 3个加标水平,测定加标回收率及其变异系数。JA和H2JA均采用ES-电离模式和MRM扫描模式,采集特征离子分别为母离子209 m/z、子离子58.7 m/z和母离子211 m/z、子离子58.7 m/z。结果表明利用UPLC-MS/MS检测的JA在2~100 ng/mL的浓度范围内呈良好的线性关系,使用氨基柱进行处理前后的变异系数分别为:10.83%和1.92%,3个加标水平的加标回收率为79.16%~93.46%。
UPLC-MS/MS quantification of endogenous jasmonic acid(JA) was carried out in rice seedlings.Analyses were conducted by UPLC-MS/MS with dihydro-JA(H 2JA) as the internal standard.JA was extracted from rice tissue with methanol,purified with solid phase extraction column(SPE-NH 2) and then followed by the UPLC-MS/MS analysis procedure.Three spiked concentration levels of 5 ng/g,20 ng/g and 100 ng/g were set for the determination of the recovery and coefficient of variation.ES-mode and MRM scan mode were used both for JA and H 2JA.Collecting characteristic ions were 209 m/z58.7 m/z and 211 m/z 58.7 m/z.The result showed that JA quantification with UPLC-MS/MS had a linear relationship in the range of 2-100 ng/mL.The coefficients of variation for SPE-NH 2 purified sample and unpurified samples were 1.92% and 10.83% respectively.The average recoveries of the three spiked concentration levels were in the range of 79.16%-93.46%.
