便塞通合剂调节慢性便秘大鼠血清GAS、MTL和SP含量实验研究 被引量:15

摘要 目的:证实便塞通合剂对便秘模型大鼠的通便作用是通过调节血中GAS、MTL和SP等胃肠激素的分泌来实现的这一假说。方法:84只SD大鼠随机分为7组:空白组、模型组、阳性对照1组(麻仁丸组)、阳性对照两组(莫沙比利组)、便塞通合剂治疗高、中、低剂量组。用复方地芬诺酯复制便秘大鼠模型,确定模型建立后,各组分别给药,每天灌胃1次,7天为1疗程,疗程间停药1天,2个疗程后,收股动脉血采用ELISA法检测。结果:便塞通高、中剂量组和莫沙比利组大鼠的GAS、MTL和SP的含量显著增高,与模型组和空白组大鼠比较差异有极显著性(P<0.01);便塞通低剂量组和麻仁丸组大鼠的GAS和MTL含量与模型组大鼠比较有统计学差异(P<0.05);各给药组组间比较可见:便塞通高、中剂量组和莫沙比利组大鼠的GAS和MTL的含量高于便塞通低剂量组和麻仁丸组大鼠的含量,有统计学意义(P<0.05)。结论:便塞通合剂能提高大鼠血浆中GAS、MTL和SP等胃肠激素的含量,而具有通便作用,并且便塞通的通便作用皆呈一定的量效依赖关系,高、中剂量组作用优于低剂量组。 Objective:To confirm this hypothesis that the function of Biansetong mixture on constipation rats was regulated by gastrin(GAS),motilin(MTL),substance P(SP).Methods:84 SD rats were randomly divided into seven groups:normal group,model group,Positive control group 1(MaRen Pills group),positive control group 2(mosapride group),high-dose BianSeTong mixture,middle-dose BianSeTong mixture and slight-dose BianSeTong mixture.Constipation rat model was established by Compound Diphenoxylate.After the success of modeling,every groups were given relevant theraperutic drugs respectively.Drugs were applied one time per day for 2 weeks.After a course of treatment(7 days),the drugs were discontinuated 1 day.All animals were killed after 2 weeks.Then femoral artery blood were detected by enzyme linked immunosorbent assay(ELISA).Results:The amount of GAS,MTL,SP in BianSeTong mixture groups it increased especially in middle-dose,high-dose group and mosapride group.They were significantly higher than model group and normal group(P0.01);The amount of GAS,MTL In Maren Pills group and the low-dose BianSeTong mixture group was higher than model group(P0.05).The amount of GAS,MTL in BianSeTong mixture middle-dose groups,high-dose group and mosapride group were significantly higher than that of Maren Pills group and the low-dose BianSeTong mixture group(P0.05).Conclusion:Biansetong mixture can increase the amount of GAS,MTL and SP in the blood of constipation rat model.It's function can depend on dose,and high-dose BianSeTong mixture,middle-dose BianSeTong mixture were super to low-dose BianSeTong mixture group.
