摘要
目的:从雷公藤根(Tripterygium wilfordii)茎及叶中分离鉴定内生真菌,并对其发酵产物进行体外抗肿瘤活性筛选。方法:采用微生物纯化方法分离雷公藤植物的内生真菌,根据菌落形态和显微特征对菌株进行分类鉴定。选用HCT-8(人结肠癌细胞)、Bel-7402(人肝癌细胞)、BGC823(人胃癌细胞)、A549(人肺癌细胞)为指标,采用MTT法对发酵产物进行体外抗肿瘤活性筛选。结果:从雷公藤不同器官中分离得到64株内生真菌,经形态观察鉴定为3目,5科,11属。其中5株内生真菌发酵产物对不同肿瘤细胞具有显著选择性细胞毒作用。结论:雷公藤内生真菌种类多样,具有很好的研究和应用价值。
Objective: to isolate the endophyte of Tripterygium wilfordii and screen its antitumor activities.Methods: endophytic fungi were indentified by morphology.Four tumor cell lines(HCT-8,Bel-7402,BGC823,A549) were selected in the MTT assay.Results: sixty-four strains of endophytic fungi were isolated from the healthy root,stem and leaf of T.wilfordii,which belonged to eleven genera,five families,and three orders.Five of them exhibited significant cytotoxic activities toward these tumor cell lines in the MTT assay.Conclusion: the endophytic fungi in T.wilfordii were diversity and rich,showing important value of development.
